Osteogenesis imperfecta, also known as brittle bone disease, is an inherited disorder that causes fragile bones and frequent fractures. This thesis shows that the disease affects not only the long bones, but also the craniofacial skeleton, including the jaw, skull, and facial bones. These abnormalities can affect chewing, speech, appearance, and dental treatment. The research demonstrates that people with osteogenesis imperfecta have an increased risk of fractures in the craniofacial region. Preclinical studies further show that inhibition of sclerostin antibody can stimulate bone formation in craniofacial bone and improve dental implant anchorage, even in weakened bone. These findings provide new opportunities for personalized regenerative treatments aimed at improving oral health, function, and quality of life for people with osteogenesis imperfecta.

Hsiao Hsin Sung Hsieh was born in Taiwan in 1975 and grew up in Chile. She earned her DDS, specialty training in Oral and Maxillofacial Surgery, and MSc from the Universidad de Chile. Since 2013, she has worked at the University of Michigan School of Dentistry, where she is currently Clinical Associate Professor.
