dr. Kevin NeumannIn recent years, there has been a growing interest in the chemical synthesis of peptides for therapeutic applications. The formation of aspartimide is one the largest impediments to the synthesis of numerous peptides. To overcome this limitation, we reported the use of cyanoylides as remarkably stable and polar protecting groups for carboxylic acids during solid phase peptide synthesis that completely prevent the formation of aspartimides. We demonstrated the potential of this new protecting group by successfully employing it for the chemical synthesis of different low-density lipoprotein class A repeats (LA-modules), which were otherwise chemically inaccessible.
