Theme 2 colloquium: "Diffusive and tethered protein-protein complexes" (Lecture)

Erik Zuiderweg Michigan June2017Protein - protein interactions are often thought to give rise to rigid complexes. However, in the field of the Hsp70 molecular chaperones, the rules seem to be different. Using NMR spectroscopy, we found evidence for a tethered complex between the Hsp70 and the ubiquitin ligase CHIP, a diffusive complex for Hsp70 and its Hsp40 co-chaperone,  as well as for Hsp70 and its autophagy clients. In the colloquium I  will give a brief introduction in the cellular function the Hsp70 system, and will explain in detail the biophysical interpretation of the NMR data that lead to these discoveries. If time permits, I will hark back to my Nijmegen PhD work on a diffusive complex of hemoglobin and a polyphosphate.
