Generation of genetically modified zebrafish

The zebrafish is extremely suitable for in vivo functional characterization of gene function and gene variants. Within the facility, we have ample experience in establishing gene knock-out and (humanized) knock-in models using CRISPR/Cas-mediated genome editing. We have multiple microinjection setups allowing for RNA-guided CRISPR/Cas-mediated targeted mutagenesis, and established a highly-efficient pipeline for the development and genetic evaluation of CRISPR-engineered zebrafish models. We are also experienced with generation of transgenic zebrafish lines using the Tol2- and PhiC31-integrase gene-delivery systems. With these tools, we can swiftly induce the expression of genetic therapy constructs or cell-specific fluorescent reporter proteins in zebrafish.
